Modulator Proteins

Unless indicated otherwise, all proteins below are human recombinant and supplied per vial as indicated in 50 ml 50 mM Tris-HCl, pH 7.0 buffer containing 14 mM b-mercaptoethanol, 1 mM benzamidine, 0.1 mM PMSF, 1 mM EDTA, 0.1% Brij-35 and 10% glycerol. I1aPP2A
GLO140 (250 ng, $285)

Also termed PHAP-1 and p33. Highly expressed in some cancers and may also be involved in regulating apoptosis. Undergoes phosphorylation in cells. Purified preparations of this oncoprotein inhibit all forms of PP2A tested to date (GLO130-132, protein phosphatases) in a substrate-selective manner. For example, the purified preparations inhibit PP2A potently (ki ~ 0.1 nM) with MBP (GLO126, kinase substrates) and histone H1 but not with casein as substrate. In contrast, the purified preparations stimulate PP1 (GLO133, protein phosphatases) in a substrate-selective manner. Purity > 90% by SDS-PAGE. Apparent Mr ~ 30-kDa.

I2aPP2A
GLO141 (250 ng, $285)

Also termed SET, PHAP-II, and TAF-1b. Highly expressed in Wilms tumor. Associates with HRX leukemia-associated fusion proteins. Occurs as fusion protein with the nucleoporin CAN in acute undifferentiated leukemia. May be involved in apoptosis. Undergoes phosphorylation in cells. Purified preparations of this oncoprotein inhibit all forms of PP2A tested to date (GLO130-132, protein phosphatases) in a substrate-selective manner. For example, the preparations inhibit PP2A potently (ki ~ 0.1 nM) with MBP (GLO126, kinase substrates) and histone H1 but not with casein as substrate. In contrast, the pruified preparations stimulate PP1 (GLO133, protein phosphatases) in a substrate-selective manner. I2aPP2A also prevents inhibition of E-CDK-2 by p21Cip1, associates with B cyclin, regulates histone binding to DNA, transcriptional activity and chromatin condensation. Purity > 90% by SDS-PAGE. Subunit A of Protein Phosphatase 2A NEW!

The A (also termed PR65) subunit of PP2A (GLO130-GLO132, protein phosphatases) serves as a scaffolding molecule to coordinate the assembly of the catalytic subunit and a variable regulatory B subunit, generating functionally diverse heterotrimers of the phosphatase. The A subunit also serves to regulate the polycation response of PP2A. Mutations of the A subunit are associated with lung and colon tumors. DEK oncoprotein NEW!

DEK is a nuclear phosphoprotein of apparent Mr ~ 43 kDa that is fused to the nucleoporin CAN as a result of a recurrent (6;9) chromosomal translocation in a subset of acute myeloid leukemias and myelodysplastic syndrome. DEK has also been identified as an autoimmune antigen in juvenile rheumatoid arthritis and other rheumatic diseases. DEK changes the topology of DNA by introducing constrained positive supercoils into protein-free and chromatin-associated DNA. DEK also associates with the transcriptional co-repressor Human Daxx, the SRm160 splicing coactivator, and the HIV-2 enhancer. DEK exhibits some structural similarity to I2aPP2A (GLO141, this page), which also occurs as a fusion protein with the nucleoporin CAN in acute undifferentiated leukemia. However, whether DEK and I2aPP2A act via similar mechanisms in vivo is currently unknown.
