Principle:
UNIQ-10 spin columns
and 96-well plates utilize silica-gel membrane technology
adsorbing selectively up to 10 µg PCR DNA or DNA fragments in
the presence of specialized binding buffers. Nucleotides,
enzymes, mineral oil and other impurities do not bind to the
membrane. PCR products or DNA fragments can be eluted with
elution buffer in small volume.
Application:
- Recovery of PCR
products from PCR reaction mixture.
- Removal of
nucleotides and primers (<50-mers).
Features:
- Rapid and
economical. Entire procedure takes 15-20 minutes (UNIQ-10) resp.
40 minutes (UNIQ-96 well).
- High yields
(60-90%). Recovers 60 bp - 40 kb DNA fragments.
- Efficient removal
of contaminants. Purified DNA can be used in any downstream
applications such as sequencing, labeling, restriction enzymatic
digestions, ligations, hybridizations or transformations.
- No phenol /
chloroform extraction or ethanol precipitation.
Procedure:
The kit uses a simple
bind-wash-elution procedure. PCR products are mixed with the
appropriate binding buffer and pass through to the column. DNA
binds to the silica-based membrane. Impurities such as enzymes,
salts, nucleotides and primers ( < 50-mer) are washed away. Pure
DNA is eluted in a small volume of the low-salt elution buffer
or water, ready for use in any downstream applications.
