Mucins belong to a family of glycoproteins which contribute to the protective and lubricating functions of mammalian mucus. Mucin glycoproteins are either membrane-associated (MUC-1) and function in cell-cell interactions or they are secreted and constitute the major macromolecular component of mucus in the gastrointestinal, respiratory and reproductive tracts. Recently, by molecular technology a number of Mucin genes have been identified [MUC-1 to MUC-9]. Signet’s MUC-1 (BC-2) antibody recognizes the breast cancer associated mucin encoded by the MUC-1 gene. The epitope recognized by the antibody has been mapped to a 5 amino acid sequence, APDTR, within the core protein. In normal tissues, MUC-1 is expressed in the apical plasma membrane of the epithelial component of many organs including breast, salivary gland, esophagus, stomach, pancreas, bile ducts, lung, kidney, bladder, and uterus. In the corresponding carcinomas, increased MUC-1 expression with altered glycosylation has been shown. High levels of MUC-1 expression have also been associated with greater invasiveness. Several studies have shown a correlation between the expression of MUC-1 and the prognosis of breast carcinoma. Other carcinomas such as lung, urogenital and gastrointestinal tumors may also be stained with the MUC-1 antibody.
