The SIGNET anti-Macrophage Antigen monoclonal antibody, clone LN5, stains the cytoplasm of human macrophages and derived tumors. In normal tissues LN5 reacts strongly with sinus histiocytes, mantle zone B cells, interdigitating reticulum, and spermatogonia. A weak reaction may be observed with kidney epithelial tubules, breast ductal epithelium, and chief cells of the stomach. In tumor tissues LN5 detects true histiocytic lymphomas, histiocytosis-X, malignant histiocytosis, and benign lymphoid lesions such as sinus histiocytosis, tuberculosis, sarcoidosis, dermatopathic lymphadenopathy, persistent generalized lymphadenopathy and Gaucher's disease.
