The Signet CD50 (186-2G9) antibody reacts
with the single chain 120kDa polypeptide cellular adhesion molecule
(CD50 antigen ICAM-3). The protein is heavily glycosylated and resistant
to treatment with phosphatidylinosital phospholipase C and therefore
probably not Pl-anchored to the membrane. CD50 antigen is constitutively
expressed at high levels on leukocytes, including epidermal, dentritic
Langerhans cells and is released from activated lymphocytes and
neutrophils. It is detected on blood vessel walls within some lymphomas
and a soluble form is delectable in blood. The CD50 molecule mediates
adhesion between leukocytes and appears to be important in the initial
interactions between T cells and dentritic cells leading to T cell
activation. In general, CD50 antigen expression is restricted to
haematological neoplasms with a tendency to be lost in high grade
lymphomas and Reed-Sternberg cells. Endothelial cells from
tumor-associated neovascularisation in both lymphoid and solid tumors
also express CD 50 antigen.
