The Signet CD23 (BU38) monoclonal antibody
recognizes the B cell differentiation antigen CD23, a 45-50kDa type II
trans-membrane glycoprotein. The antigen acts as a low affinity IgE
receptor (FcER2) and is expressed at variable degrees on follicular
mantle B cells and also occasionally in the splenic marginal zones or in
germinal centers. The CD23 antibody gives a characteristic-binding
pattern with follicular dentritic cells but the antigen is absent on
plasma cells and B cell progenitors. Expression of CD23 antigen is
greatest in activated B-lymphocytes. Cleavage of the membrane bound form
releases a series of soluble fragments, which still bind the IgG except
for the 12 kDa moiety. This antibody also reacts with B cells in chronic
lymphocytic leukemia and some cases of centroblastic/centrocytic
lymphoma.
